METHODOLOGY
REC ASSAY

The Rec Assay is used to determine the direct DNA-damaging capacity of the samples. Recombinant-proficient (H17) and recombinant-deficient (M45) strains of Bacillus subtilis are grown overnight in B2-broth. Each culture is streaked radially on the dry surface of the broth agar. A paper disk saturated with the sample is placed over the starting points of the streaks. The plates are kept at 4-5°C for 24 hours, followed by incubation at 37°C for about 20 hours. Zones of inhibition are measured.
